Retinal abnormalities in transgenic mice overexpressing aberrant human FUS[1-359] gene

چکیده

Retinal damage is an optional sign in a number of neurodegenerative diseases, including amyotrophic lateral sclerosis (ALS). The aim this work was to assess the structural and functional state retina murine model ALS caused by overexpression aberrant FUS protein [1-359]. retinal examination carried out on 12 transgenic 13 wild-type mice 2.5–3 months age. study revealed not statistically significant higher level ophthalmoscopic violations FUS[1-359] mice. Moreover, gene expression assay confirmed increased inflammatory genes Vegfa, Il1b, Il6, Icam1, Tnfa. However, despite detected abnormalities, western blot analysis quantitative PCR did detect mRNA products transgene

Transgenic mice overexpressing reticulon 3 develop neuritic abnormalities.

Dystrophic neurites are swollen dendrites or axons recognizable near amyloid plaques as a part of important pathological feature of Alzheimer's disease (AD). We report herein that reticulon 3 (RTN3) is accumulated in a distinct population of dystrophic neurites named as RTN3 immunoreactive dystrophic neurites (RIDNs). The occurrence of RIDNs is concomitant with the formation of high-molecular-w...

Hyperlipidemia and cutaneous abnormalities in transgenic mice overexpressing human apolipoprotein C1.

Transgenic mice were generated with different levels of human apolipoprotein C1 (APOC1) expression in liver and skin. At 2 mo of age, serum levels of cholesterol, triglycerides (TG), and FFA were strongly elevated in APOC1 transgenic mice compared with wild-type mice. These elevated levels of serum cholesterol and TG were due mainly to an accumulation of VLDL particles in the circulation. In ad...

Galanin overexpressing transgenic mice.

Galanin overexpressing transgenic mice (GAL-tg) were generated on two different promoters. Both lines of GAL-tg displayed high levels of galanin in the hippocampus and reduced sensitivity to seizures, as compared to their respective wildtype littermate controls (WT). Performance deficits on learning and memory tasks, impaired long-term potentiation, reduced hippocampal excitability, lower evoke...

Combined hyperlipidemia in transgenic mice overexpressing human apolipoprotein Cl.

We have generated transgenic mice over-expressing human apolipoprotein CI (apo CI) using the native gene joined to the downstream 154-bp liver-specific enhancer that we defined for apo E. Human apo CI (HuCI)-transgenic mice showed elevation of plasma triglycerides (mg/dl) compared to controls in both the fasted (211 +/- 81 vs 123 +/- 52, P = 0.0001) and fed (265 +/- 105 vs 146 +/- 68, P < 0.000...

Endotoxemia in transgenic mice overexpressing human glutathione peroxidases.

In response to endotoxemia induced by administration of lipopolysaccharide, a complex series of reactions occurs in mammalian tissues. During this inflammation response, cells produce different mediators, such as reactive oxygen species, a number of arachidonic acid metabolites, and cytokines. The reactive oxygen species thus generated have been suggested to produce tissue injury as a result of...
